Description:
The adenosine metabolism pathway involved in CD73 plays an important role in TME mediated immune escape. CD73 is an extracellular 5 '- nucleotidase encoded by the NT5E gene, with a molecular weight of approximately 70 kDa. It is anchored to the outer surface of the cell membrane by glycosyl phosphatidylinositol (GPI) to form homologous dimers, or cleaved and free in the circulatory system. CD39 upstream of CD73 can catalyze ATP to produce adenosine monophosphate (AMP), which can be converted to adenosine by CD73. Adenosine binds downstream adenosine receptors (A2AR or A2BR), which activate protein kinases PKA and CSK while inhibiting a series of immune activation related signaling pathways such as MAPK and PKC, thus exerting a wide range of immunosuppressive effects.
