Description:
CTLA-4, also known as CD152, is the first immune checkpoint protein successfully targeted by ipilimumab. CTLA-4 is a transmembrane glycoprotein that binds to CD80 (B7-1) and CD86 (B7-2) proteins on APCs. CTLA-4 acts as a ‘off’ switch when bound to its ligands. PD-1, also known as CD279, primarily regulates effector T cell activity within tissues and tumors, rather than regulating T cell activation in lymphoid organs. PD-1 is a cell surface receptor that inhibits T cell proliferation and cytokine production (IFN-γ, IL-2, etc.), and reduces T cell survival by interacting with its ligands PD-L1 and PD–L2.
